The optimization of conditions to promote the stemness of pluripotent cells in vitro is instrumental for their use in advanced therapies. We show here that exposure of human iPSCs and human ESCs to low concentrations of the chemical NO donor DETA/NO leads to stabilization of hypoxia-inducible factors (HIF-1α and HIF-2α) under normoxia, with this effect being dependent on diminished Pro 402 hydroxylation and decreased degradation by the proteasome. Moreover, the master genes of pluripotency, NANOG and OCT-4, were upregulated. NO also induces a shift in the metabolic profile of PSCs, with an increased expression of hypoxia response genes in glycolysis. Furthermore, a reduction in the mitochondrial membrane potential with lower oxygen consumption and increased expression of mitochondrial fusion regulators, such as DRP1, was observed. The results reported here indicate that NO mimics hypoxia response in human PSCs and enhances their stemness properties when cultured under normoxic condi...

ABSTRACT Recently, some well-known univariate aging classes of lifetime distributions have been characterized by means of properties of their quantile functions and excess-wealth functions. The generalization of the univariate aging notions to the multivariate case involve, among other factors, appropriate definitions of multivariate quantiles or regression representation and related notions, which are able to correctly describe the intrinsic characteristic of the concepts of aging that should be generalized. The multivariate versions of these notions, which are characterized by using the multivariate u-quantiles and the multivariate excess-wealth function, are considered in this paper. Relationships between such multivariate aging classes are studied, and examples are provided.

ABSTRACT Univariate stochastic and dispersive ordering have extensively been characterized by many authors over the last two decades. Stochastic orderings are also applied in economic. In particular, it is interesting to compare situations where one utility function (or one distribution function) is obtained from the other by means of some operation that has an economic meaning. To this end, stochastic properties for distributions associated to normal distribution in sampling are studied. An application of the multivariate dispersion order in the problem of detection and characterization of influential observations in regression analysis is also shown. This problem can often be associated to compare two multivariate t-distributions.

ABSTRACT Linear models constitute the primary statistical technique for any experimental science. A major topic in this area is the detection of influential subsets of data, that is, of observations that are influential in terms of their effect on the estimation of parameters in linear regression or of the total population parameters. Numerous studies exist on radiocarbon dating which propose a value consensus and remove possible outliers after the corresponding testing. An influence analysis for the value consensus from a Bayesian perspective is developed in this article.

Background Most proteins have evolved in specific cellular compartments that limit their functions and potential interactions. On the other hand, motifs define amino acid arrangements conserved between protein family members and represent powerful tools for assigning function to protein sequences. The ideal motif would identify all members of a protein family but in practice many motifs identify both family members and unrelated proteins, referred to as True Positive (TP) and False Positive (FP) sequences, respectively. Results To address the relationship between protein motifs, protein function and cellular localization, we systematically assigned subcellular localization data to motif sequences from the comprehensive PROSITE sequence motif database. Using this data we analyzed relationships between localization and function. We find that TPs and FPs have a strong tendency to localize in different compartments. When multiple localizations are considered, TPs are usually distributed...

ABSTRACT The test for exponentiality of a dataset in terms of a specific aging property constitutes an interesting problem in reliability analysis. To this end, a wide variety of tests are proposed in the literature. In this paper, the excess-wealth function is recalled and new asymptotic properties are studied. By using the characterization of the exponential distribution based on the excess-wealth function, a new exponentiality test is proposed. Through simulation techniques, it is shown that this new test works well on small sample sizes. The exact null distribution and normality asymptotic is also obtained for the statistic proposed. This test and a new empirical graph based on the excess-wealth function are applied to extreme-value examples.

Consider two random vectors X1 and X2 whose distributions are defined according to the multivariate frailty approach, and let Xk,t=[Xk−t|Xk>t], k=1,2, be the corresponding vectors of residual lifetimes at . Conditions for multivariate stochastic comparisons of random vectors described by the frailty approach have been recently presented in Misra et al. (2009). Here we prosecute their study, providing sufficient conditions for the stochastic comparison X1,t≤stX2,t, where t is an arbitrary vector in Rn. Sufficient conditions for the stochastic comparisons Xi,t≤stXi,t+v, where t is as above and v is a vector with non-negative components, are presented too.

Consider two vectors X1 and X2 of random lifetimes whose distributions are dened via the frailty approach, and let Xk;t = (Xk tj Xk > t), k = 1; 2, be the corresponding vectors of residual lifetimes at t = (t1;:::;tn);ti 2 R+;i = 1;:::;n. Here we describe sucient conditions for the stochastic comparison X1;t st X2;t for every vector t of non-negative times.
